Objective:We investigated the potential effect of Orlistat on lipase production in liver. Liver-specific enzyme activities of lipase and substrate were assayed by a method that is based on the inhibition of lipase by the drug. We also measured the effect of Orlistat on the expression of the gene encoding hepatic lipase. In addition, the influence of Orlistat on the expression of hepatic lipase was evaluated by real-time quantitative reverse transcriptase-PCR (qRT-PCR).

Methods:Total RNA was extracted from the cultures of three liver-specific enzyme-inducing strains (L>[+]L), L>[+]L/KL/KL, and L>[+]L>[+]L/KL/KL. The total RNA of the three strains was subjected to RT-PCR with the specific primers L>[+]L>[+]L/KL/KL/KL. The results of qRT-PCR were analyzed by using the ABI 7500 Real-Time PCR System (Applied Biosystems) and the relative expression of the target gene was normalized with respect to the negative control. All experiments were performed in triplicate. The results were analyzed by using the Student's t-test.
